Retatrutide is an investigational triple receptor agonist engineered to simultaneously activate GIP, GLP-1, and glucagon receptors. This novel triple mechanism represents an evolution beyond dual incretin agonism, incorporating glucagon receptor activation to drive hepatic energy expenditure and fatty acid oxidation. The 5mg research-grade lyophilized formulation (≥98% purity) is intended exclusively for laboratory investigation. The addition of glucagon receptor agonism provides a distinct third axis of action. While GIP and GLP-1 activation address insulin secretion, appetite suppression, and glucose-dependent effects, glucagon receptor stimulation directly increases energy expenditure through enhanced hepatic fatty acid oxidation and thermogenesis. This triple approach yields weight loss outcomes that consistently exceed those of dual agonists in comparative research models. Phase 2 clinical data demonstrate up to 24.2% mean body weight reduction at 48 weeks at the highest dose, surpassing results seen with tirzepatide in analogous study populations. Additional research endpoints include marked improvements in HbA1c, liver fat content (of particular interest in non-alcoholic steatohepatitis research), blood pressure, and atherogenic lipid profiles. The compound is currently in Phase 3 development with Eli Lilly and is not yet FDA-approved. This material is supplied for laboratory research use only — including receptor pharmacology studies, metabolic pathway investigations, and preclinical research applications. Not for human or veterinary use. As an investigational compound not approved by regulatory agencies, it is restricted to in vitro and animal research settings.
